Análisis integrado de morfología y movilidad espermática humana con el uso del sperm class analyzer
The aim of this work was validated the use of Sperm Class Analyzer® (SCA) in the human clinic laboratory, so to start on a integrated analysis of the cinematic and morphometric parameteres of spermatozoa. Previously, the suitable conditions of image acquisition were determined. Analysis were made with diluted and no diluted semen, positive and negative contrast, and 10x and 20x objetive (9 samples). The best work condition was no diluted semen, negative contrast and 20x objetive. Firstly, manual sperm counting was compared between Makler chamber and Neubauer hemocytometer (56 samples), showing high correlation. Beside, subjetive and objetive determinations were correlated, so cinematic parameters (VSL, VAP, WOP, LIN, STR, ALH and BCF) among them. Finally, principal components were calculated and analyzed. The understanding of results was based in the dominat factors so in the rate of explicative total variance. Eigth parameters were reduced to three principal components. On another hand, a morphometric study was parallel realized. Equally, subjetive and objetive determinations were correlated, so different morphometric parameters (lenght, width, area, perimeter, acrosome rate, Ellipticity, Rugosity, Elongation and Regularity of head; width and area of middle-piece; and distance between larger axises of head and middle-piece) among them. Lastly, principal components were calculated and analyzed. Thirteen morphometric parameters were reduced to three principal component. To finish, cinematic and morphometric parameters were integrated, studing their correlation and reducing the total of variables to five principal components: shape of head, size of head, index, velocity and middle-piece. In conclusion, the use of SCA is valited to spermatic motility and morphology analysis and the reduction to principal components introduces stearier criterion in seminal assessment. Finally, to assess if actual diagnostic criterions were deducible of new obtained variables, seminal samples were classified according to the WHO guidelines (1992, 1999). Five diagnostic groups were obtained. Cinematics and morphometrics parameters and principal components were compared between the groups, no showing correlation between actual diagnostic criterion and cuantitative parameters. Therefore, the definition of new diagnostic parameters more precise is necessary.
