Perfil protÃ©ico da membrana externa da Leptospira interrogans sorovariedade hardoprajitino.
Outer membrane proteins (OMPs) were isolated from Leptospira interrogans serovar hardjoprajitno using Triton X114, precipitated with acetone, and separated by electrophoresis 2-D and SDS PAGE and resolved by western blot. Gel migration patterns were compared usingimage analysis of 35 gel-purified proteins. Among the proteins identified by this approach included the outer membrane lipoproteins ipL22/Loa22, LipL32, Omp34, LipL41 and LipL63. The SDS PAGE identified 14 protein profiles in total. The major leptospiral OMPs were 63 KDa (LipL63), 47 KDa (LipL48), 29 KDa (LipL32) and 22 KDa (LipL22/Loa22). The results by western blot showed that antiserum from experimentally infected rabbits and naturally infected bovine evidentiated different protein patterns. The outer membrane proteins of 51,1 KDa, 36 KDa, 27KDa, 24,5 KDa, 20,5 KDa, 17,7 KDa and 14 KDa were principally detected by rabbit antibody. In contrast, two leptospiral OMPs of 47 KDa (Omp45) and 29 KDa (LipL32) were detected in significantly greater amounts using bovine serum. These findings suggested differences asdetermined by patogenicity and protein structure of L. interrogans serovar hardjoprajitno.
